The Proteas are still seeking their first series win of the summer after suffering a hefty defeat in the deciding T20I by the Australians at Newlands on Wednesday.
Janneman Malan will make his debut in the 50-over format for the Proteas after being added to the squad in the week, while Temba Bavuma returns after missing the recent T20 series due to injury.
Kyle Verreynne will also earn his first Protea cap with Faf du Plessis and Rassie van der Dussen being rested for the series.
The Proteas were struck a big blow on Friday when fast bowler Kagiso Rabada was ruled out for the series and the upcoming one to India with a groin problem.
Australia have called up South African-born Marnus Labuschagne for the ODIs, and he will be looking to carry his recent Test form into the shorter format.
